After returning from vacation, I powered my computer on. I received the THREATS DETECTED. How do I know if they've been removed? What action should I be taking. ALSO, how can I update my definitions manually.

Welcome to the Webroot Community
If Webroot detected a threat then it will remove these threats to Quaratine and the Webroot will rescan to make sure these threats are gone.
Would you check whether anything has been moved to Quarantine? To do that click on the gear/cog symbol to the right of the PC Security tab, and on the panel displayed as a result click on the 'Quarantine' tabe. If there is anything recorded there?

Webroot doesn't use any definitions and works 100% cloud based.
There's a huge cloud database where all threat information is stored and as soon as a new type of malware is added your already protected. So as long as your client is connected to the Internet you're always up to date and software updates are also silently installed, which means that you don't have to take any actions.
